Light producing animal model (LPTA®) Crl:CD-1 (ICR)BR-Tg(Cyp1a2-luc)-Xen, commonly called Cyp1a2-luc, carries an 8.4 kb mouse cytochrome p450 1a2 (Cyp1a2) promoter, a native intron 1 (1067 bp), and modified firefly luciferase cDNA (Promega pGL-3). Basal expression of the reporter is highest in liver and signals in males were much higher than in females. Injections of Cyp1a2 inducers such as 2,3,7,8-tetrachlorodibenzo-p-dioxin, 3-methylcholanthrene, phenobarbital, etc., resulted in a time-dependent induction of luciferase expression in liver. The responses to aryl hydrocarbon compounds are similar to a humanized AhR transgenic animal line, but much less than those in the Ah responsive strains such as C57BL/6. The Cyp1a2-luc LPTA® animal model is useful in studying the kinetics of the Cyp1a2 gene regulation by xenobiotics, Cyp1a2-mediated drug-drug interactions, and toxicity in the outbred mouse strain.
